Can I replace my aging, full size 24-patch color target with a SpyderCHECKR 24?

Yes, if you are using a 24-patch target for visual functions, the SpyderCHECKR 24 should be large enough and similar enough to replace it. If you are using your 24-patch target with third party software then you should be able to continue to do so, though you will need to flip the patch order of every second row before processing the target image, to achieve correct patch reading order. You can flip the patch order in your editing application before importing into the third party software. If you wish to use the SpyderCHECKR software for camera calibration instead of your older 24 patch target, the results from the SpyderCHECKR software should be as good, or better, than what you have achieved in the past.
