Spyder4TV wants me to eliminate all ambient light during the calibration, but I don't usually watch TV in these lighting conditions. Will this effect the overall picture when I restore my room lighting back to normal?

The Spyder4TV calibration process is designed to operate in optimal lighting conditions to preform accurately and consistently.

It is important that you always do a visual assessment or "fine-tune" your TV using the additional test patterns.
This will allow you to set your room lighting to your desired level, then use the additional test patterns from the Tool menu.
